CYP2C9 - Test for the administration of Siponimod
CYP2C9 genetic testing determines a patient's CYP2C9 genotype before treatment with siponimod. CYP2C9 is a major enzyme involved in siponimod metabolism, and genetic variation can affect drug exposure and the recommended maintenance dose. Genotyping therefore provides clinically relevant information to support appropriate siponimod use and dosing.
Contraindications to siponimod include patients homozygous for the CYP2C9*3/*3 genotype. They may have significant arrhythmias or conduction disorders (unless the patient has got a pacemaker) and with myocardial infarction, angina, stroke, decompensated heart failure or class III or IV heart failure.
