Scopul studiului a fost de a exploata poten\u021bialul unui panel multigen pentru analiza muta\u021biilor \u00een genele c\u0103ii recombin\u0103rii omoloage (HR), precum \u0219i a pierderii heterozigozit\u0103\u021bii (LOH), ca predictori ai r\u0103spunsului la tratament.<\/p>\n\n\n\n<p>Muta\u021bii \u00een genele BRCA1\/2 au fost detectate la 3,09% dintre pacien\u021bi, \u00een timp ce muta\u021bii \u00een alte gene HR au fost g\u0103site \u00een 10,65% din cazuri. \u00cen cancerele de prostat\u0103, ovarian, de s\u00e2n \u0219i pancreatic cu inhibitori PARP aproba\u021bi disponibili, ratele de detectare a muta\u021biilor HR au crescut semnificativ: 40% (14\/35), 26,83% (22\/82), 15,85% (13\/82) \u0219i 10% (14\/140) respectiv.<\/p>\n\n\n\n<p>\u00cen plus, valoarea %LOH a fost calculat\u0103 \u0219i la 201 pacien\u021bi. Valoarea median\u0103 a LOH ob\u021binut\u0103 a fost de 19,22% (min 0%, max 99%), cu 88 de probe (43,78%) prezent\u00e2nd o valoare LOH mai mare dec\u00e2t mediana \u0219i 113 (56,22%) probe cu valoare mai mic\u0103 dec\u00e2t mediana. Au fost observate rate crescute de LOH pentru cancerele de s\u00e2n (68,18%) \u0219i ovarian (52,63%). \u00cen timp ce \u00een cancerul de prostat\u0103 \u0219i pancreas procentul de probe cu rate ridicate de LOH a fost mai mic, 0% \u0219i, respectiv, 35,71% din tumori. Muta\u021biile HR au fost foarte asociate cu %LOH ridicat la pacien\u021bii cu cancer ovarian, deoarece 50% din cazurile cu o muta\u021bie a genei HR cu risc ridicat (BRCA1\/2, PALB2, RAD51C) au avut valori crescute ale LOH, \u00een timp ce 22% din cazuri \u00een cazul celor cu risc sc\u0103zut. Popula\u021bia LOH, purtat\u0103 o muta\u021bie \u00eentr-o gen\u0103 HR cu risc sc\u0103zut (RAD51B, FANCA).
